Бета-амилоид снижает синтез АТФ митохондриями клеток нейробластомы человека

Аннотация
Известно, что пептид бета-амилоид (Aβ), являющийся ключевым в патогенезе болезни Альцгеймера и образующий токсичные олигомерные формы, влияет на функцию митохондрий. В представленной работе с использованием люциферин-люциферазной реакции показаны изменения синтеза АТФ митохондриями, выделенными из клеток нейробластомы человека SH‑SY5Y после их культивирования в присутствии мономерного Aβ в наномолярной концентрации. Уровень синтеза АТФ измеряли в присутствии субстратов одного из комплексов дыхательной цепи – I, II и IV – и соответствующих ингибиторов других комплексов. Показано, что Aβ значительно уменьшает скорость продукции и количество АТФ, синтезируемого митохондриями. Установлено, что влияние Aβ на синтез АТФ не обусловлено непосредственным действием на комплексы дыхательной цепи I, II, IV. Полученные результаты указывают на возможные причины митохондриальной дисфункции в нейронах при болезни Альцгеймера.
