Структурные особенности интермедиата репарации нуклеосомной ДНК влияют на активность PARP2 в процессе HPF1‑независимой автомодификации

Аннотация

ДНК-зависимые ферменты клеточного ядра поли(ADP‑рибоза)-полимеразы 1 и 2 (PARP1 и PARP2) вовлечены в регуляцию различных путей репарации ДНК, включая эксцизионную репарацию оснований. Эти ферменты активируются при связывании с повреждениями ДНК и синтезируют отрицательно заряженный полимер – поли(ADP‑рибозу) (poly(ADP-ribose), PAR), ковалентно связанный с аминокислотными остатками белков-мишеней, в том числе самих PARP. Природа повреждений ДНК влияет на активность PARP2: фермент эффективнее активируется на разрывах ДНК, фланкированных фосфатом. Однако на сегодняшний день неясно, какие стадии реакции авто‑PAR-илирования PARP2 наиболее зависимы от структуры повреждённой ДНК. В данной работе мы исследовали зависимость активности PARP2 на различных стадиях реакции автомодификации (проводимой в отсутствие фактора PAR-илирования гистонов HPF1) от наличия однонуклеотидной бреши и её положения в ДНК, в том числе в составе нуклеосомы. Полученные результаты позволяют предположить, что наличие бреши влияет на сродство фермента к ДНК/нуклеосоме, определяющее число каталитически активных молекул PARP2 и эффективность инициации PAR-илирования. В то же время локализация специфичного повреждения в структуре ДНК/нуклеосомы, а именно удалённость от тупых концов ДНК и окружение гистоновых хвостов влияет на процесс удлинения PAR. Таким образом, структура повреждённой ДНК может оказывать влияние на количество и размер полимера, синтезируемого PARP2.
