Нарушение транспортёров стеринов семейства Lam приводит к дефектам споруляции у дрожжей Saccharomyces cerevisiae

Аннотация

Основная роль стеринов в клетке – поддержка барьерной функции плазматической мембраны (ПМ), поэтому их концентрация в этом компартменте максимальна среди всех клеточных мембран. У дрожжей Saccharomyces cerevisiae транспорт стеринов между различными мембранными компартментами клетки обеспечивается белками семейств Osh и Lam. Считается, что белки Lam1–Lam4 осуществляют пассивный транспорт стеринов из ПМ в мембраны эндоплазматического ретикулума (ЭР). Белки Lam5–Lam6 переносят стерины в местах контактов ЭР с вакуолями и митохондриями. Делеция генов семейства LAM не снижает скорость роста клеток в стандартных условиях, поэтому биологическая роль этих белков остаётся не до конца ясной. В данной работе высказывается предположение, что белки семейства Lam могут играть роль в спорообразовании дрожжей, поскольку плазматическая мембрана спор формируется de novo из везикул, образованных мембранами ЭР, менее насыщенных стеринами, чем ПМ, что обусловливает потребность в транспорте стерина в новообразующуюся ПМ спор. Для проверки этой гипотезы были получены диплоидные штаммы, в которых в гомозиготном состоянии были делетированы гены LAM1LAM4 и LAM5LAM6. Было показано, что одновременная делеция генов LAM5LAM6 снижает как долю асков в культуре, так и количество спор в каждом аске. В то же время одновременная делеция генов LAM1LAM4 приводила к снижению доли полных асков, но не ингибировала инициацию спорообразования. Также было обнаружено, что делеция генов LAM1LAM4 приводит к утолщению клеточной стенки и появлению дефектов в её строении. У поверхности клеточной стенки таких спор были обнаружены кластеры осмиофильных везикул. Споры с делецией генов семейства LAM демонстрировали сниженную резистентность к тепловому шоку и щёлочи. В совокупности наши данные указывают на то, что транспорт стеринов белками семейства Lam важен для нормального протекания процесса спорообразования.
