Особенности эпителиально-мезенхимального перехода, вызываемого драйверными мутациями сигнального пути PI3K/AKT в эпителиальных клетках молочной железы

Аннотация

Сигнальный путь PI3K/AKT является одним из важнейших в клетке и вовлечен в регуляцию пролиферации, выживания и миграции клеток. Мутации участников PI3K/AKT-пути часто связаны с прогрессией и метастазированием злокачественных опухолей. Начальным этапом метастазирования является эпителиально-мезенхимальный переход (ЭМП), при котором опухолевые клетки приобретают способность к миграции. Литературные данные о влиянии драйверных мутаций PI3K/AKT-пути на миграционные способности опухолевых клеток противоречивы. Мы рассмотрели изменение подвижности и ЭМП эпителиальных клеток молочной железы под влиянием четырех драйверных мутаций, затрагивающих разные уровни регуляторного пути PI3K/AKT и часто выявляемых при развитии злокачественных опухолей. Анализ подвижности клеток, экспрессии маркеров ЭМП и морфологии межклеточных контактов показал, что все эти мутации приводят к инициации неполного ЭМП, так как во всех изученных линиях сохраняется экспрессия Е‑кадгерина и клетки сохраняют межклеточные контакты. Мы проанализировали типы ЭМП, вызванных разными мутациями, и показали, что ускорение миграции не определяется степенью прогрессии ЭМП по направлению к мезенхимальному фенотипу. Так, наибольшее увеличение миграционных способностей клеток отмечено при мутации PIK3CA H1047R/+, которая приводит к наиболее выраженному мезенхимальному фенотипу, и при нокауте PTEN, который сохраняет наиболее эпителиальный фенотип. Анализ показал, что наибольшее влияние на ЭМП и клеточную подвижность оказывают те мутации, которые опосредованно влияют на активность регуляторного пути MAPK/ERK и усиливают активацию ERK.
