Аннотация

Вирус гепатита B инфицирует гепатоциты человека, вызывая острую либо хроническую инфекцию печени. При хроническом течении инфекции продолжающаяся репликация вируса способствует прогрессирующему повреждению ткани печени, что со временем может приводить к развитию цирроза или гепатоцеллюлярной карциномы. Одним из перспективных направлений противовирусной терапии является активация ферментов-цитидиндезаминаз семейства APOBEC/AID, которые способны вызывать мутационную деградацию вируса гепатита B. В рамках предложенного нами подхода, основанного на контролируемой экспрессии APOBEC с применением системы транскрипционной активации CRISPRa и модифицированных sgRNA, мы исследовали противовирусные и онкогенные эффекты активации генов, кодирующих APOBEC3C, APOBEC3D и APOBEC3H.
