Ингибирование протеасомы как побочный эффект эверолимуса, специфичный к типу рака

Аннотация
Эверолимус, ингибитор mTORC1, может влиять на активность протеасомы аналогично бортезомибу, что требует дальнейшего исследования. В работе мы использовали сверхбыструю экспрессионную протеомику в сочетании с анализом жизнеспособности клеток и активности протеасомы для выявления потенциальных вторичных мишеней эверолимуса и получения более полного понимания механизма его действия в протеомах линий злокачественных клеток. Результаты по эверолимусу сравнивались с результатами, полученными для бортезомиба и лонидамина, которые использовались в качестве положительного и отрицательного контроля ингибирования протеасомы соответственно. Наши результаты показывают, что эверолимус ингибирует 20S протеасому в клетках рака легких (A549) и толстой кишки (HCT116), при этом не оказывая заметного эффекта в клетках рака молочной железы (MCF‑7). Была построена in silico модель взаимодействия эверолимуса с 20S протеасомой, предполагающая аллостерический механизм ингибирования.
