Усиление биогенеза лизосом при воздействии доксорубицина на клетки HCT116 в условиях нормоксии

Аннотация
Гидрофобные слабоосновные препараты, такие как доксорубицин и сунитиниб, являются в настоящее время важными компонентами химиотерапии опухолевых заболеваний. Показано, что ряд таких соединений индуцирует увеличение общего объёма лизосом в опухолевых клетках. Известно также, что гипоксия, характерная для опухолевых клеток in vivo, приводит к химиорезистентности за счёт изоляции доксорубицина в лизосомах. Для усиления эффективности химиотерапии предлагаются разные подходы, в том числе направленные на дисрегуляцию лизосом. Признаётся, что блок аутофагии часто является способом снижения химиорезистентности. Необходимо показать, существует ли, наоборот, влияние доксорубицина на физиологию лизосом. В настоящей работе на клетках карциномы толстой кишки HCT116 показано, что доксорубицин локализуется в значительной степени в лизосомах даже при нормоксии. При нормоксии доксорубицин вызывает увеличение суммарного объёма лизосом, тогда как в условиях дефицита кислорода подобный эффект слабее. Совместное применение доксорубицина и хлорохина, широко известного лизосомотропного препарата, приводит к увеличению объёма лизосом как в нормоксии, так и в гипоксии. В условиях нормоксии доксорубицин активирует фактор биогенеза лизосом TFEB, что может объяснять увеличение суммарного объёма данных органелл. Также показана функциональная активность лизосом в присутствии доксорубицина. Эффект увеличения лизосом и усиления их деградационной активности в присутствии доксорубицина также показан для линии фибросаркомы HT1080. Суммируя, впервые показано влияние доксорубицина на параметры лизосом в разных кислородных условиях на моделях опухолевых клеточных линий.
