С-Концевой фрагмент филамина С, содержащий 19–24 иммуноглобулин-подобные домены, избирательно взаимодействует с малым белком теплового шока HspB7

КЛЮЧЕВЫЕ СЛОВА: филамин С, малые белки теплового шока, α‑кристаллиновый домен, иммуноглобулин-подобные домены.

Аннотация

Филамин С является адаптерным белком, участвующим в регуляции цитоскелета и взаимодействующим более чем с 90 белками-партнёрами, среди которых выделяют группу малых белков теплового шока. Механизм взаимодействия этих белков с филамином остаётся не полностью изученным. В данной работе с использованием иммунохимических методов, а также методов гель-фильтрации, нативного электрофореза и химического «сшивания» было проанализировано взаимодействие длинного С‑концевого фрагмента, содержащего 19–24 иммуноглобулин-подобные домены (Ig) филамина С человека (FLNC19-24), с различными малыми белками теплового шока. Все использованные методы свидетельствуют о том, что из пяти исследованных малых белков теплового шока (HspB1, а также его 3D мутантная форма, имитирующая фосфорилирование, HspB5, HspB6, HspB7 и HspB8) только HspB7 был способен образовывать комплексы c FLNC19‑24. Тот факт, что HspB7 взаимодействует как с изолированным 24‑м Ig, так и с фрагментами филамина, содержащими 22–24 и 19–24 Ig, убедительно свидетельствует о прочном взаимодействии HspB7 с филамином С и позволяет подтвердить локализацию участка взаимодействия двух белков. Селективное связывание α‑кристаллинового домена HspB7 в области 24‑го домена филамина С вызывает диссоциацию его димеров, предположительно способствуя ускоренному перемещению филамина С в клетке и репарации повреждений сократительного аппарата.
