Внутренний промотор и сайты связывания микроРНК в кодирующей области: особенности регуляции эволюционно молодого гена lawc у Drosophila melanogaster

Аннотация
Влияние микроРНК (миРНК) на экспрессию консервативных генов хорошо известно, однако нет сведений об их вкладе в контроль работы эволюционно молодых генов. Ген lawc возник de novo в 5′‑нетранслируемой области гена Trf2. Мы использовали модельную систему этих генов с целью оценить возможную роль миРНК как инструмента эволюционной адаптации, обеспечивающего согласованную работу генов в условиях их перекрытия. Исследование включало применение трансгенных линий дрозофил с индуцибельной экспрессией предшественников миРНК (miR‑4968, miR‑2491, miR‑13b‑1), а также созданного GFP‑сенсора, позволяющего визуализировать подавление экспрессии целевых последовательностей под действием миРНК in vivo. Для тканеспецифической активации трансгенов использовали дрожжевую двухкомпонентную систему Gal4/UAS. Люциферазный тест на культурах клеток Drosophila S2 и человека HEK293 применяли для подтверждения функциональности уникального промотора, предсказанного внутри кодирующей области (CDS) гена lawc, и оценки влияния миРНК на его экспрессию в клетках S2. Дополнительно проводили иммуноблоттинг, ОТ‑ПЦР и конфокальную микроскопию для анализа уровня белка и мРНК, а также визуализацию активности GFP-сенсора в тканях дрозофилы. Полученные данные позволили экспериментально подтвердить функциональность внутреннего промотора гена lawc и роль исследуемых миРНК схожего эволюционного возраста в модуляции его экспрессии. На примере гена lawc мы заключили, что промоторы в составе CDS характерны не только для генов микроорганизмов и дрожжей, но могут встречаться и у эволюционно молодых генов высших эукариот, возникших в регуляторной зоне другого гена. Примечательно, что эти гены быстро приобретают регуляцию миРНК схожего эволюционного возраста, что указывает на ко-эволюционный механизм, лежащий в основе формирования новых регуляторных сетей, обеспечивающих адаптацию двух генов в условиях их перекрытия.
