Координация нуклеозидтрифосфатов в активном центре терминальной дезоксирибонуклеотидилтрансферазы человека при матрица-независимом синтезе ДНК

Аннотация

Терминальная дезоксирибонуклеотидилтрансфераза человека (TdT) – ДНК-полимераза семейства X, отвечает за повышение разнообразия иммуноглобулинов и генов T‑клеточных рецепторов в процессе V(D)J‑рекомбинации путем матрица-независимого присоединения случайных нуклеотидов к 3′‑концу перестраиваемых сегментов V‑D и J‑D. Согласно рентгеноструктурным данным, координация входящего dNTP и 3′‑концевых нуклеотидов одноцепочечного праймера в активном центре TdT осуществляется за счет контактов с гидрофобными (W449, F404 и L397) и гидрофильными (R336, H342, D395, E456, R453 и R457) остатками, формирующими dNTP-связывающий карман. При этом остатки R336, H342 и D345 принимают непосредственное участие в ориентации фосфатных групп входящего dNTP и стабилизации 3′‑концевого нуклеотида ДНК-праймера. В настоящей работе был выполнен анализ последствий замены данных остатков методами молекулярной динамики и экспериментальной проверки ферментативных свойств мутантных форм TdT человека, содержащих замены R336Q, H342A и D345E. Полученные данные показали, что замена H342A вызывает увеличение константы диссоциации комплекса фермента с dNTP и ДНК-праймером и значительно уменьшает эффективность присоединения dNTP к растущей цепи. Замена D345E нарушает координацию ионов металла-кофактора и также оказывает влияние на эффективность каталитической реакции. Замена остатка R336 приводит к значительной дестабилизации структуры белковой глобулы и полной потере каталитической активности фермента. Полученные данные позволили установить молекулярные причины влияния замен R336Q, H342A и D345E на эффективность каталитического присоединения dNTP к ДНК-праймеру. Функциональное картирование активного центра нуклеотидилтрансфераз в перспективе может быть использовано для проведения рационального дизайна мутантных форм фермента, востребованных для реализации молекулярно-биологических и биотехнологических приложений.
