Молекулярные механизмы феномена антитело-зависимого усиления вирусной инфекции

Аннотация

Антитело-зависимое усиление инфекции (ADE) – явление, которое наблюдается при вторичной инфекции, вызываемой некоторыми вирусами, прежде всего из семейства Flaviviridae, в том числе такими, как вирус лихорадки денге (DENV), вирус Зика (ZIKV), вирус Западного Нила (WNV), вирус клещевого энцефалита, вирус жёлтой лихорадки и некоторые другие. Различают две разновидности ADE: опосредованное Fc‑рецепторами и рецепторами к компонентам комплемента. В первом варианте в ADE участвуют 4 вида Fc‑рецепторов к молекуле IgG: FcRI, FcRIIa, FcRIIb и FcRIIIa, которые обеспечивают связывание с клеткой-мишенью комплекса вируса с антителом и дальнейшую интернализацию этого комплекса. Fc-опосредованное поглощение вируса является активным процессом, в котором принимает участие целый ряд сигнальных молекул и запускаются разветвлённые каскады активационных событий. Fc-опосредованный путь входа вируса увеличивает вирусную нагрузку и переводит клетку с антивирусного ответа на провоспалительный путь реакции, что способствует более эффективной репликации вируса. В отличие от других Fc-рецепторов, молекула FcRIIb при связывании с IgG ингибирует захват комплекса вирус-антитело и предотвращает ADE. Крайне нежелательным является развитие ADE в ответ на вакцинацию. Понимание биохимических механизмов, определяющих ADE, имеет важное значение при разработке новых вакцин, а также терапевтических моноклональных антител, применяемых для защиты от вирусных инфекций.
