О механизме селективного химического замещения бактериофеофитинов в реакционных центрах Rhodobacter sphaeroides R‑26

Аннотация

С целью выяснения механизма сайт-селективного химического замещения хромофоров в реакционных центрах (РЦ) фотосинтетических бактерий внешними пигментами исследована эффективность встраивания растительного феофитина a (Фео) в сайты связывания молекул бактериофеофитина a (БФео) в изолированных РЦ Rhodobacter sphaeroides R‑26 в зависимости от температуры среды инкубации, агрегационного состояния Фео и присутствия органического растворителя (ацетона). Степень селективного замещения фотохимически неактивных молекул БФео HB составляла ~15% при инкубации смеси РЦ/Фео при 5 °C в условиях, когда Фео находился в водно-детергентной среде инкубации в виде мономеров в составе свободных мицелл детергентов. Эффективность замещения увеличивалась до 40% при инкубации при 25 °С и составляла при этой температуре около 100%, если в среду инкубации добавлялся 10%‑ный ацетон. Степень пигментного замещения была примерно в 2 раза выше при обеих температурах, когда в качестве инкубационной среды использовалась смесь мономеров и димеров Фео в присутствии 10%‑ного ацетона. Удаление ацетона из такой среды с сохранением пигментных форм приводило к значительному снижению эффективности встраивания Фео. Влияние ацетона на пигментное замещение наблюдалось также при повышенной температуре инкубации (43,5 °С), когда частично замещались функционально активные молекулы БФео HA. Результаты обсуждаются в рамках механизма, согласно которому (1) температурно-зависимые внутренние движения белка РЦ являются фактором, облегчающим высвобождение молекулы БФео из сайта связывания и одновременное внедрение молекулы Фео в тот же сайт в сопряжённом процессе, (2) роль температурного фактора в значительной степени зависит от стерической доступности связывающих белковых карманов, (3) встраивание Фео происходит из пула мономерных молекул, включённых в мицеллы РЦ‑детергент, (4) присутствие ацетона в среде инкубации облегчает обмен мономерными молекулами Фео между мицеллами в растворе и детергентным поясом РЦ.
